Analysis of restriction fragment length polymorphism in the kappa-casein gene related to weight expected progeny difference in Nellore cattle Genet. Mol. Biol.
Biase,Fernando H.; Garnero,Analia Del V.; Bezerra,Luiz A.F.; Rosa,Arthur J.M.; Lôbo,Raysildo B.; Martelli,Lúcia.
Restriction fragment length polymorphism (RFLP) has been detected at the bovine kappa-casein locus. The polymorphism has been analyzed for its effects in cattle production, mostly for milk traits and even for maternal effect on pre-weaning weights. We used polymerase chain reaction - restriction fragment length polymorphism (PCR-RFLP) to genotype 408 Nellore animals for the non-silent mutation (Thr/Ile136 and Asp/Ala148) that characterizes the A and B variants of the polymorphism and compared expected progeny difference (EPD) for a maternal effect on 120 and 210 days weights and direct EPD for 120, 210, 450 and 550 day weight between AA and AB animals. The EPD values were obtained from the University of São Paulo (Brazil) Nellore Cattle Breeding Program,...

Development and characterization of microsatellite loci of Microglanis cottoides (Siluriformes: Pseudopimelodidae) and cross-species amplification Neotropical Ichthyology
Souza-Shibatta,Lenice; Ferreira,Dhiego Gomes; Oliveira,Claudio; Almeida,Fernanda Simões de; Shibatta,Oscar Akio; Sofia,Silvia Helena.
Thirteen microsatellite loci were isolated and characterized in Microglanis cottoides. Of these, two were monomorphic and 11 were polymorphic. These polymorphic loci tested on 24 individuals from a wild population produced a total of 108 different alleles, with levels of variability high, ranging from 2 to 20, with an average of 8.3 alleles per locus. The observed and expected heterozygosity ranged from 0.125 to 0.958 and from 0.119 to 0.931, respectively. A high combined probability of paternity exclusion value and a low probability combined genetic identity value obtained show that the set of loci described herein displays good suitability for paternity studies and differentiation of M. cottoides. Additionally, all thirteen microsatellite primers...

Development of mussel-specific expression vectors suitable for transgenic organisms: implication on the establishment of mollusc continuous cell lines ArchiMer
Delsert, Claude; Cancela Da Fonseca, Leonor.
The main objective of this project was to develop the tools for the in-depth understanding of mollusc biology and pathology. Such a goal has been reached in other biological systems through the development of molecular biology and cell culture. Indeed, development of a mollusc cell culture system is an essential step to approach many of the problems related to mollusc pathology, in particular to use molecular biology tools to study viral infection of these organisms. No continous cell line is available from bivalves and while cell cultures have been obtained from these organisms they are difficult to develop and labor intensive, survival cultures being maintained only for a few weeks. Our goal has been to study in detail many of the parameters involved in...

Importance and future of individual markers for the ecosystem approach to fisheries ArchiMer
Fromentin, Jean-marc; Ernande, Bruno; Fablet, Ronan; De Pontual, Helene.
The use of genetic, biochemical and electronic markers in population biology and ecology has been growing tremendously during the last two decades. The first part of this paper aims at reviewing the main principles and advances of these individual markers through a few key applications on exploited marine fish populations. The second part is more prospective and investigates some possibilities that could arise in the near future through: (i) the development of new markers, (ii) the combination of different markers and (iii) the combination of quantitative approaches-whether classical or new-with individual markers. It is finally stressed how crucial individual markers will be to unravel the biocomplexity of wild fish populations and the key role they...

Mode de reconnaissance hôte symbionte en milieux extrêmes : cas du modèle symbiotique Rimicaris exoculata ArchiMer
Le Bloa, Simon.
Deprived of light, the deep-sea hydrothermal vents are extremes ecosystems sustained by micobial chemosynthesis. These environments are colonized by animal species living in close relationships with these chemoautotrophic micro-organisms, eating them or establishing long term interactions with them, may they be trophic or not only. The shrimp Rimicaris exoculata is an endemic hydrothermal species of the Mid-Atlantic Ridge (MAR) sites. This crustacean represents the predominant macrofauna of some sites of the MAR. It lives in symbiotic association with two distinct microbial communities qualified as ectosymbiosis. One is located in its gill chamber and one in its gut. First, this work focused on the study of bacterial communication (Quorum Sensing or QS)...

PCR survey of 50 introns in animals: Cross-amplification of homologous EPIC loci in eight non-bilaterian, protostome and deuterostome phyla ArchiMer
Gerard, K.; Guilloton, E.; Arnaud-haond, Sophie; Aurelle, D.; Bastrop, R.; Chevaldonne, P.; Derycke, S.; Hanel, R.; Lapegue, Sylvie; Lejeusne, C.; Mousset, Sylvain; Ramsak, A.; Remerie, T.; Viard, Frederique; Feral, Jean-pierre; Chenuil, A..
Exon Primed Intron Crossing (EPIC) markers providemolecular tools that are susceptible to be variable within specieswhile remaining amplifiable by PCR using potentially universal primers. In this studywe tested the possibility of obtaining PCR products from 50 EPIC markers on 23 species belonging to seven different phyla (Porifera, Cnidaria, Arthropoda, Nematoda, Mollusca, Annelida, Echinodermata) using 70 new primer pairs. A previous study had identified and tested those loci in a dozen species, including another phylum, Urochordata (Chenuil et al., 2010). Results were contrasted among species. The best results were achieved with the oyster (Mollusca) where 28 loci provided amplicons susceptible to contain an intron according to their size. This was...

Polymorphism of the grasshopper Rhammatocerus schistocercoides populations revealed by RAPD PAB
Silva,João Batista Tavares da; Tigano,Myrian Silvana; Magalhães,Bonifácio Peixoto; Cordeiro,Célia Maria Torres.
The objective of this work was to study the genetic variability of the grasshopper Rhammatocerus schistocercoides (Orthoptera: Acrididae) using RAPD analysis among individuals from three populations, one from Colombia and two from Brazil (Goiás and Mato Grosso States). Ninety scorable binary markers were obtained by fingerprinting with 11 oligonucleotide primers. Most of the polymorphism was attributed to 42 markers with variable frequency among the different populations. Although the existence of significant difference among populations (P<0.0001), most of the genetic variability was found within populations (87.7% of total variation). Pairwise distances between Colombian and Brazilian populations were 0.12 (P<0.0001) and 0.18 (P<0.0001) for...

Santi Pinthukas; Christoph Knorr; Ratchaneewan Kumphakam; Bertram Brenig; Kesinee Gatphayak.
Disease is a major problem in the swine production. The study of genes associated with disease resistance characteristics is used to select for innate immunity in swine. The Mx1 gene was found to have potent antiviral affect against some virus and can be hereditary. The study of Mx1 polymorphisms in among 3 pig breeds (Thai native pig breeds, Chinese pig breeds and European pig breeds) were found SNPs in exon 8 (c. 1022C>G; 1078A>C) that has allele frequencies of statistical significance between the breeds. (P<0.05) The two variant in exon 15 found silent point mutation (c. 2120G>T) และ 11-bp indels (c.2117_2128 indels CGGCGCCGGCT) that has allele frequencies of statistical significance in some breed (P<0.05)especially in German Landrace and...
